Abstract summary
The two hemispheres of the human brain differ functionally and structurally. Despite over a century of research, the extent to which brain asymmetry is influenced by sex, handedness, age, and genetic factors is still controversial. Here we present the largest ever analysis of subcortical brain asymmetries, in a harmonized multi-site study using meta-analysis methods. Volumetric asymmetry of seven subcortical structures was assessed in 15,847 MRI scans from 52 datasets worldwide. There were sex differences in the asymmetry of the globus pallidus and putamen. Heritability estimates, derived from 1170 subjects belonging to 71 extended pedigrees, revealed that additive genetic factors influenced the asymmetry of these two structures and that of the hippocampus and thalamus. Handedness had no detectable effect on subcortical asymmetries, even in this unprecedented sample size, but the asymmetry of the putamen varied with age. Genetic drivers of asymmetry in the hippocampus, thalamus and basal ganglia may affect variability in human cognition, including susceptibility to psychiatric disorders.Authors & Co-authors: Guadalupe Mathias vanErp Whelan Zwiers Abe Abramovic Agartz Andreassen Arias-Vásquez Aribisala Armstrong Arolt Artiges Ayesa-Arriola Baboyan Banaschewski Barker Bastin Baune Blangero Bokde Boedhoe Bose Brem Brodaty Bromberg Brooks Büchel Buitelaar Calhoun Cannon Cattrell Cheng Conrod Conzelmann Corvin Crespo-Facorro Crivello Dannlowski de Zubicaray de Zwarte Deary Desrivières Doan Donohoe Dørum Ehrlich Espeseth Fernández Flor Fouche Frouin Fukunaga Gallinat Garavan Gill Suarez Gowland Grabe Grotegerd Gruber Hagenaars Hashimoto Hauser Heinz Hibar Hoekstra Hoogman Howells Hu Hulshoff Pol Huyser Ittermann Jahanshad Jönsson Jurk Kahn Kelly Kraemer Kugel Kwon Lemaitre Lesch Lochner Luciano Marquand Martin Martínez-Zalacaín Martinot Mataix-Cols Mather McDonald McMahon Medland Menchón Morris Mothersill Maniega Mwangi Nakamae Nakao Narayanaswaamy Nees Nordvik Onnink Opel Ophoff Paillère Martinot Papadopoulos Orfanos Pauli Paus Poustka Reddy Renteria Roiz-Santiáñez Roos Royle Sachdev Sánchez-Juan Schmaal Schumann Shumskaya Smolka Soares Soriano-Mas Stein Strike Toro Turner Tzourio-Mazoyer Uhlmann Hernández van den Heuvel van der Meer van Haren Veltman Venkatasubramanian Vetter Vuletic Walitza Walter Walton Wang Wardlaw Wen Westlye Whelan Wittfeld Wolfers Wright Xu Xu Yun Zhao Franke Thompson Glahn Mazoyer Fisher Francks
